  • Pbx1b, Pbx3b, and Pbx4 proteins containing a 6xHis affinity tag at their N terminus were expressed in E. coli SG13009 [pREP4] induced with 0.5 mM IPTG for 3 hr at 30 C. Soluble His-tagged Pbx proteins were purified under native conditions using Ni-NTA agarose (Qiagen) and were used to immunize one rabbit (150 ug of recombinant protein per injection, initial immunization with Pbx3b, first boost with Pbx4, second boost with Pbx1b, 3rd boost with Pbx3b and Pbx4). The pan-Pbx rabbit antiserum was further affinity purified using GST-Pbx3b coupled to cyanogen bromide-activated Sephrose 4B(Sigma). The affinity-purified pan-Pbx antibodies detect four zebrafish Pbx proteins (Pbx1, Pbx3, Pbx4, and Pbxy [now named Pbx2] ) equally well on a Western blot (data not shown).
